大鼠视网膜表达生长抑素mRNA细胞的发生 被引量:1

摘要 目的 :研究视网膜中表达生长抑素mRNA神经元的发生和发育规律。方法 :原位杂交组织化学。结果 :胚胎第十三天 (E1 3 ) ,视网膜内层就可以检测到生长抑素mRNA阳性细胞。从E1 4到E1 7阳性细胞增加迅速 ,E1 7达到高峰 ,此时视网膜节细胞层大部分为生长抑素mRNA阳性细胞 ,细胞排列紧密。从E1 8开始 ,阳性细胞逐渐减少 ,细胞排列开始稀疏。出生后当 (PND0 )阳性细胞数显著下降。从PND0到PND1 5 ,阳性细胞继续减少 ,阳性细胞主要位于节细胞层。PND2 0时 ,阳性细胞的数目及分布均与成年动物相似 ,部分已迁移至内核层的内表面。细胞开始或停止表达生长抑素mRNA均是从视网膜中央开始 ,然后向周围。结论 :首次发现生长抑素基因在胚胎期的节细胞中一过性表达 ,提示生长抑素在视网膜节细胞的产生 ,分化以及视觉通路的形成与成熟有关。 To study the ontogeny of neurons-expressing somatostatin mRNA in the rat retina. Methods: In situ hybridization histochemistry was used. Results: Neurons-expressing SOM mRNA in the retinas of fetuses became detectable at E13. From E14 to E17, the number of neurons-expressing SOM mRNA increased rapidly and reached the peak value at E17. At E17 most of the cells in the developing ganglion cell layer expressed SOM mRNA and they were arranged very closely. From E18 the positive cells arranged sparsely gradually, and the number decreased. At the postnatal day 0(PDN0) the positive cells decreased dramatically. From PND0 to PND15, the number of the positive cells in the retinas decreased gradually and the positive cells were still located in the developing ganglion cell layer. At PND20, distribution pattern and the number of the positive cells were similar to that of adult, the positive cells migrated into inner nuclei layer. Conclusion: The transient presence of SOM in most of the ganglion cells in the developing ganglion cell layer suggests that there is a possible involvement of SOM in the generation and differentiation of the ganglion cells, formations and maturation of retina-visual center projections.
