日本血吸虫31kDa组织蛋白酶B DNA疫苗保护性免疫效果观察 被引量:10

摘要 目的 观察日本血吸虫 31k Da组织蛋白酶 B DNA疫苗 (Sj31B1 N)在小鼠体内的保护性免疫效果。方法 用 Sj31B1 N和 Sj31B1 N +p UC DNA疫苗肌注免疫 BAL B/ C小鼠 ,用空白质粒载体VR10 12做对照 ,在 0、4、8周共免疫 3次 ,每次免疫前及末次免疫后 4周从尾静脉采血收集血清 ,经EL ISA法检测小鼠血清抗体升高时 ,用日本血吸虫尾蚴攻击感染小鼠。结果  Sj31B1 N和 Sj31B1 N+p U C减虫率分别为 2 4.2 %和 2 2 .0 % ;肝卵减少率分别为 34 .9%和 39.5 % ,每雌肝卵减少率分别为 30 .4%和 35 .3%。结论 小鼠接种 Sj31B1 N后对日本血吸虫感染有减虫、减卵作用及抗雌虫生殖的作用。加质粒佐剂 p Objective To study protective immunity effect in mice induced by Schistosoma japonicum cathepsin B DNA vaccine (Sj31B 1N). Methods BALB/c mice were vaccinated intramuscularly with the nucleic acid(Sj31B 1N), the gene encoding cathepsin B of Schistosoma japonicum. Each group was immunized three times at week 0,4,8. Mice vaccinated with VR1012 blank vector served as negative control. Mice were challenged with the cercariae when specific antibody in serum was detected by ELISA. Sera from vaccinated mice were collected from the tail vein at week 0,4,8,12, respectively. Results Vaccination with Sj31B 1N and Sj31B 1N+pUC resulted in significant protection with 24 2% and 22 0% reduction in worm burden, 39 5% and 34 9% reduction in liver tissue eggs, 35 3% and 30 4% reduction in liver tissue eggs per female worm, respectively. Conclusion Sj31B 1N DNA vaccine can induce a certain level of specific protective immunity in mice against infection of S. japonicum and anti fecundity of female worm. However, pUC as an adjuvant didn't enhance the protective immunity in this experiment.
