摘要
目的 :探讨芍药苷对PC12细胞钙超载损伤模型的保护作用及其机制。方法 :采用组织培养法 ,以大鼠肾上腺嗜铬细胞瘤克隆株PC12细胞为材料 ,制备氯化钾及N 甲基 D 门冬氨酸诱导的钙超载损伤模型。结果 :形态学检查发现芍药苷对 2种不同类型的钙超载损伤模型中PC12细胞均具有明显保护作用 ,MTT法活细胞测定提示 12 .5~ 2 0 0 μg·mL-1的芍药苷可显著提高损伤模型中PC12细胞存活数 ,减少胞内乳酸脱氢酶渗漏 ,并显著减少胞内钙离子浓度。结论 :芍药苷对钙超载所致PC12细胞损伤有显著的保护作用。
Objective:To investigate the protective effect of paeoniflorin (PAE) on calcium overloading injury models and its mechanism.Methods:Two injury models induced by potassium chloride(KCl) and N-methyl-D-aspartate(NMDA) were used to measure the action of PAE on PC12 cells.Results:Morphological examination revealed that PAE possessed obvious protective effects on PC12 cells in injury models.MTT and LDH measurement in supernate indicated that PAE in dose of 12.5~200μg·mL -1 increased significantly the survival number of PC12 cells in injury models and reduced significantly the Ca 2+ level in cytoplasm.Conclusion:PAE possesses protective effect on two calcium overloading injury models in cultured PC12 cells.
