摘要
目的:探讨病毒性心肌炎心肌离子泵功能及钠钙含量变化,并用卡托普利进行干预。方法:雄性Balb/c小鼠随机分为柯萨奇B3病毒(CVB3)感染组(IG)、CVB3感染加卡托普利治疗组(ICG)及对照组(CG)。在不同病程分别测定心肌Na+,K+-ATPase、Ca2+-ATPase活性,心肌钠钙含量及组织病理变化。结果:与CG比较,IG心肌Na+,K+-ATPase、Ca2+-ATPase活性明显下降(P<0.01),钠钙含量显著升高(P<0.01),并早于心肌结构变化。ICG上述各项指标较IG显著改善(P<0.05),心肌炎症、坏死也明显减轻(P<0.05)。结论:心肌离子泵功能下降,反常性Ca2+超载可能是造成病毒感染后心肌继发性损伤的重要因素;卡托普利能明显保护病毒性心肌炎心肌离子泵功能,降低心肌钠钙含量,改善心肌组织结构,促进预后。
Objective:To explore the changes of myocardial ion ic pump functions and sodium and calcium concentrations in mice with viral myoca rditis,and to observe the intervention effects of captopril on them.Method s:The male Balb/c mice were randomly divided into three groups:infected c oxsackie B3 virus (CVB3)group (IG), infected CVB3 and treated with captopr il group (ICG) and control group (CG). The activities of myocardial Na+,K+-A TPase and Ca2+-ATPase,the concentrations of myocardial sodium and cation a s well as the myocardial pathologic changes were investigated in different perio ds. Results:As compared to CG,the activities of myocardial Na+,K +-ATPase and Ca2+-ATPase of IG declined (P<0.01), the concentrations of myocardial sodium and calcium of IG increased(P<0.01),and these changes were observed prior to myocardial histologic abnormalities.All of these changes were obviously improved in ICG(P <0.05),the myocytes necrosis and the myocytes inflammatory cells infiltration were reduced(P<0.05). Conclusion: The inhibiti on of ionic pump functions and Ca2+ overload in Captopril can protect myo cardial ionic pump functions, reduce concentrations of myocardial sodium and ca lcium,ameliorate the myocardial histologic structure and improve the prognos is in viral myocarditis.
