摘要
目的 从本实验室已建立的 Ig G类抗日本血吸虫抗原不同表位的单抗中筛选抗独特型抗体 NP30的独特型抗体 (id,Ab1 )。方法 应用间接 EL ISA经典法 ,NP30作为抗原包板。加 Ig G各亚类杂交瘤细胞培养上清孵育 ,用辣根过氧化物酶 (HRP)标记抗鼠 Ig G捕捉。结果 单抗 NP5 6与NP30呈阳性反应 ,NP5 6的 Ig同型为 Ig G2 b。免疫组织化学显示 NP5 6与虫卵内毛蚴头腺及毛蚴膜呈阳性反应。结论 NP5 6是 NP30的特异性 id,可用于
Objective To select idiotypic antibody (id, Ab 1) of monoclonal anti idiotypic antibody NP30 of Schistosoma japonicum [WT5”BZ]from the IgG monoclonal antibodies, which are special to various epitopes of [WT5”BX]Schistosoma japonicum, [WT5”BZ]in the larboratory. [WT5”HZ]Methods [WT5”BZ]The traditional indirect ELISA was used and reactive board was coated by NP30. Then the cultural supernatant fluid of IgG monoclonal antibodies was added and captured by sheep anti mouse IgG labelled with HRP. [WT5”HZ]Results [WT5”BZ]The monoclonal antibody NP56 had positive reaction with NP30. The immunoglobulin isotype of NP56 was determined as IgG 2b . NP56 reacted positively with the head gland and membrane of miracidia in eggs of Schistosoma japonicum by the method of immunohistochemistry. Conclusion NP56 is the special id of NP30 and can be used for identification of the mimic antigen of NP30.
