摘要
在获得稳定表达HLA G分子的K5 62细胞克隆的基础上 ,观察 4h及 16h不同效靶比例时NK细胞对靶细胞的杀伤效应 ,结果显示 ,NK细胞对转染HLA G基因的K5 62细胞的杀伤作用较未转染细胞明显降低 (P <0 0 1) ,用W6/ 32及BFL 1单克隆抗体阻断HLA G分子 ,可以恢复NK细胞的杀伤活性。用Y9/Z2 70及XA185 /Z2 70单抗特异性阻断抑制性受体CD94/NKG2A 。
On the basis of a K562 cell line which steadily express the HLA G molecule,the protective role of this molecule was assessed in 4 h and 16 h 3H TdR release assay in which the effector cells were mixed wth 1×10 5/ml target cells The experimental results showed that the cytolytic activity of K562 cells transfected with the HLA G gene was significantly reduced in comparison with those cells untransfected with this gene Futhermore,this inhibitory effect could be restored by blockage of the HLA G molecule with anti HLA G BFL 1 mAb and pan class 1 W6/32,but not blocked by Y9/Z270 and XA185/Z270 mAb which were specific to the CD94/NKG2A
