摘要
目的 探讨光动力疗法 (PDT)对鼻咽癌细胞 (NPC)BaxmRNA表达和凋亡的影响。方法 体外培养的NPC细胞 ,分别在PDT作用 12、2 4、36和 48h后用电镜动态观察分析细胞凋亡现象和凋亡指数 ,同时采用逆转录 -聚合酶链反应技术 (RT PCR)检测BaxmRNA的表达情况。结果 PDT作用 12、2 4、36和 48h后NPC细胞凋亡指数分别为 6 .7%、71.6 %、86 .4%和 98.3 % ;同时BaxmRNA表达检测发现。各对照组NPC细胞仅可见BaxmRNA扩增产物 ,实验组NPC细胞BaxmRNA表达均呈阳性 ,随PDT作用时间延长而逐渐增强 ,两者结果呈现一致。结论 PDT体外可诱导鼻咽癌细胞凋亡 ,细胞凋亡指数上升与凋亡相关基因BaxmRNA表达上调有关。
Objective To cxplore the effects of photodynamic therapy(PDT) on the expression of Bax mRNA and apotosis in nasopharygeal carcinoma cell (NPC) in vitro.Methods Bax mRNA expression in NPC cell line both before and after PDT in vitro were detected by reverse transcriptase polymerase chain reaction(RT PCR) and the ultrastructure destructive process of NPC cell line after PDT by electro microscopy(TEM).Results By PDT for 12,24,36,48 h,NPC apoptosis indexes were 6.7% , 71.6% , 86.4% , 98.3% ,respectively.The expression of Bax mRNA were positive in NPC cell line before PDT,and significantly enhanced in a time dependent fashion after PDT.The change of Bax mRNA expression was concordant with the cell apoptosis rates.Conclusion PDT induce NPC cell death in vitro apoptosis,and partially may be associated with the upregulation of Bax mRNA.
