摘要
目的 :建立一种测定尿 5 ` -核苷酸磷酸二酯酶 (5′- NPD)的新方法 ,试图为临床诊断提供一项检测早期肾损害的生化指标。方法 :以 5′- [5 -碘吲哚酚 - (3) ]胸腺嘧啶核苷酸铵盐作为检测尿 5′-核苷酸磷酸二酯酶的底物 ,在 p H9.0缓冲体系中进行酶促反应后 ,在波长 A5 80 nm下测定尿 5′- NPD的吸光度 (OD)值。结果 :该法重复性 :cv =1.98% ;尿5′- NPD酶活力与 OD值的线性关系 :Y=4.116 X+0 .0 2 76 ,r =0 .993.用该法分别检测 30例肾病患者和 5 0例正常人的尿 5′- NPD,肾病患者的尿 5′- NPD酶活力明显高于正常人 (P <0 .0 1)。结论 :提示尿 5′-
Objective:To offer a biochemical parameter in clinical diagnosis of patients with impairment of renal function, a new method for detecting 5′-nucleotide phosphodiesterase (5′-NPD) in human urine was established.Method: Using 5′-(5-iodoindol-3-yl) thymidylate as a substrate of 5′-NPD, the absorbency of reacted solution was detected after enzymatic reaction. Result :The coefficients of variation was 1.98% and the linear curve was obtained (Y=4.116X+0.0276,γ=0.993) between enzymatic activity of 5′-NPD and absorbency. The result was showed that enzymatic activity of 5′-NPD in 30 patients with impairment of renal function and 50 normal human urine samples was significantly different (P<0.01). Conclusions: The results suggest that 5′-NPD in urine could be regard as a significant biochemical parameter in diagnosis of patients with impairment of renal function.
