摘要
采用细胞培养法 ,免疫组化实验技术和流式细胞仪等项技术 ,研究了bcl 2基因反义核酸和砷剂单独及联合应用对K5 6 2细胞的生物效应 ,DNA及bcl 2蛋白的变化。结果显示 ,As2 O3(2 .0 μmol L) +bcl 2ASODN(10 .0 μmol L)联合作用比单用As2 O3 (2 .0 μmol L)或bcl 2ASODN(10 .0 μmol L)显著增强诱导细胞凋亡的作用 (P <0 .0 1) ;流式细胞仪检测显示 ,联合作用引起的bcl 2蛋白表达亦明显减少 (P <0 .1)。结论表明 ,bcl 2基因反义核酸能明显提高和显著增强K5 6 2白血病细胞对As2 O3
The biological reaction, changes of cellular DNA and bcl 2 protein content of K562 cells to bcl 2 ASODN or As 2O 3 alone or bcl 2 ASODN associated with As 2O 3 were investigated by cellular microculture, immunological tissue chemistry method, and flow cytometry. The results showed that there is much more induce apoptotic effect of As 2O 3 on K562 cells while it combined with bcl 2 ASODN (ASODN 10.0 μmol/L+As 2O 3 5.0 μmol/L) than bcl 2 ASODN or As 2O 3 alone (ASODN 10.0 μmol/L or As 2O 3 5.0 μmol/L), (P<0.01), and so does inhibitory effect of bcl 2 protein expression by flow cytometry (P<0.01). In conclusions, the bcl 2 ASODN can enhance the drug sensetivity of K562 leukemic cells to As 2O 3.
