摘要
通过大量试验 ,证明双层平板法是光合细菌测数的较好方法。它弥补了传统方法的测数不准确、操作繁琐的缺点。通过正交设计试验 ,确定影响光合细菌测数的4个主要因素的最佳组合为 :底层培养基琼脂含量为0.8 % ,烤板时间为0.5h,上层培养基温度为50℃ ,上层培养基琼脂含量为1.0 %。
Lots of experiments showed that double-plate method was suitble for counting photosynthetic bacteria (PSB). It overcomes the shortage of old methods which are not accurate and convenient for operating. The four main factors affecting PSB counting were determined through changing the experimental conditions. The optimum combination is 0.8% agar in the lower pant of medium, 0.5 hour toasting plate, 50℃ for the temperature at the upper part of medium, 1.0% agar in the upper part of medium.
