摘要
目的 :探讨妊娠高血压综合征与N5 ,N10 -亚甲基四氢叶酸还原酶 (MTHFR)基因及血浆同型半胱氨酸(Hcy)水平的关系。方法 :运用多聚酶链反应 -限制性内切酶片段长度多态性技术 (PCR -RFLP)检测 42例妊高征患者 ,36例正常妊娠妇女及 30例非妊娠妇女 ,MTHFR基因多态性 ,用高效液相色谱仪和电化学检测法测定 42例妊高征患者 ,36例正常妊娠妇女及 30例非妊娠妇女血浆总Hcy水平。结果 :MTHFR基因型有3种 ,即纯合子 (T/T)型 ,杂合子 (T/C)型 ,纯合子 (C/C)型。非妊娠组 ,正常妊娠组及妊高征组之间的基因型和等位基因频率差异均无显著性 (P >0 .0 5 )。妊高征组总Hcy水平 (14.0 6± 6 .0 6 ) μmol/L明显高于正常妊娠组 (P <0 .0 0 1) ;并随妊高征病情的加重 ,总Hcy水平呈逐渐上升趋势 ,重度妊高征明显高于轻度妊高征 (P<0 .0 5 )。结论 :MTHFR基因突变与妊高征发生无关 ;
Objective:To study the relationship between pregnancy induced hypertension (PIH) and the polymorphisms of MTHFR gene and plasma homocysteine levels.Methods:30 healthy nonpregnant women,36 normal pregnant women and 42 women with PIH were recruited in the study.The polymorphisms of MTHFR gene were analyzed using PCR-RFLP technique.Their plasma homocysteine concentrations were measured by HPLC and electrochemical detection.Results:MTHFR has 3 genotypes:homozygote (T/T),heterozygote(T/C),homozygote(C/C).There was no significant differencc between the genotype and allele frequency amony the nonpregnant group,normal pregnant group and PIH group (P>0.05).Total Hcy level in PIH group was remar remarkably higher than in normal pregnant group(P<0.001).The toltal Hcy level increased with the aggrevation of the PIH.Conclusions:MTHFR mutation is irrelevant to the occurrence of PIH.High Hcy level in blood is a new risk factor in the develpoment of this disease.
