摘要
目的 :用人绒毛膜促性腺激素(hCG) ,进行ELISA竞争法测定激素的方法研究。方法 :用ELISA竞争法检测尿中hCG。以抗hCG单克隆抗体包被酶标板孔 ,先后加入含有待测hCG的尿标本及酶标记的hCG,使二者共同竞争与板孔中的单抗结合 ,经显色后按颜色深浅、OD值大小判断阴、阳性。结果 :经对临床近100份标本检测结果证明本法与乳胶法判断的结果符合率为97 %。结论 :ELISA竞争法也可用于检测激素及其它小分子抗原 ,方法简便、省时。
Objective: To observe the method for detecting hormones we used hCG with competitive ELISA. Methods: Described here is a competitive ELISA for detecting hCG in urine.Anti-hCG monoclonal antibodies recognizing different epitopes were coated in microplate wells; the urine sample and hCG-peroxidase conjugate were added to the wells in order. After OPD was added,the deep or light colors (or different OD value) showed the negative or positive resaffs respectively.Results:The results of about 100 clinical urine samples showed that the result of the method correlated well to the traditional latex agglutination inhibition test,the correlation rate is 97%.Conclusion: Competitive ELISA can also be used to determine hormones and other small molecular antigens,and it is easy and quick to perform.
