摘要
比较基因组分析证明 ,玉米和水稻基因组间存在广泛的同线性和共线性。对水稻和玉米基因的原位杂交定位可以揭示禾本科植物基因组结构的共同特点和进化规律。用与 Xa- 5连锁的 RG556及RG556筛选出的 BAC克隆 4 4 B4作探针对水稻广陆矮四号和玉米自交系黄早四进行了原位杂交 ,在水稻第 5号染色体短臂和玉米第 8号染色体长臂检出了杂交信号 ,信号与着丝粒百分距离分别为 4 8.85、4 7.35、 63.0 5。 BAC- FISH信号检出率 ( 4 1% )大大高于 RFL P探针 RG556( 8% )的检出率。 BAC- FISH需要 Cot- 1DNA的封阻。取材于稻兜的根尖经低温处理后能促进姊妹染色单体的分开 。
Comparative genome analyses have demonstrated that there were widesoread syntenies and colinearities of the genes among rice and maize. Analysis of rice and maize genome by ISH will reveal the basic evolutionary units of cereal genomes. In our study, we used RG556 and a BAC clone 44B4 which linked to the gene Xa 5 as probe in ISH. The tested plant were O.sativa subsp.indica cv.“Guang Lu Ai 4” and the inbred line of Zea mays cv. In rice the Xa 5 was located in the short arm of the chromosome 5, the percent distance from the centromere to the signal was about 48.85. In maize, one hybridized sites was detected on the long arm of the chromosome 8. It suggested that the homologous sequences of Xa 5 exist in maize. By BAC FISH technique, the detection rate of signal was over 40%, much higher than that by FISH with RFLP probe, only 8%. It demonstrated the feasibility and advantage of BAC FISH technique in gene comparative physical location. The root tip come from rice in field was dealed with 4℃ can improve the detection rate of double singal on chromosome.
