摘要
通过鸟枪法克隆了甜菜夜蛾核多角体病毒 (SeMNPV)基因组DNAEcoRⅠ酶切的 2 2kb片段 .序列分析表明 ,该片段含有gp37基因 .SeMNPVgp37基因的开放阅读框为 80 1个核苷酸 ,编码 2 6 8个氨基酸 ,预测蛋白质相对分子质量为 30 40 0 .在gp37基因起始密码子上游有一典型的杆状病毒晚期基因启动子序列ATAAG .同其它昆虫杆状病毒和昆虫痘病毒GP37/Fusolin蛋白的比较结果表明 ,SeMNPVGP37与已知gp37/fusolin基因的氨基酸序列同源性较高 (5 0 %~ 6 8% ) ,在其蛋白序列内存在类似的保守区和可能的N_连接糖基化位点 .在SeMNPVgp37基因下游存在一个完整阅读框和部分egt基因序列 .
An EcoRⅠ 2 2 kb fragment of Spodoptera exigua multicapsid nucleopolyhedrovirus (SeMNPV) genome DNA was cloned by using random cloning method.The gp37 gene of SeMNPV which is contained in this fragment has been identified.The open reading frame of the SeMNPV gp37 gene is 801 nucleotides long encoding a putative protein of 268 amino acids with an Mr of about 30 400.The 5′-nonencoding region upstream of a translational start site contains one late consensus motif (ATAAG).The SeMNPV gp37 gene has 50%~68% amino acid identity to other baculovirus gp37 genes and entomopoxvirus fusolin genes. Conserved regions of GP37/Fusolin very high amino acid conservation and potential N-linked glycosylation sites were also identified in Se\%MNPV GP37 sequence.
