nNOS在缺血性老年大鼠海马及皮层神经元中的表达及超微结构变化 被引量:2

nNOS expression and morphological study in hippocampus and cerebral cortex neuron of aged rats with cerebral ischemia

摘要 目的 研究神经型一氧化氮合酶 (neuronalnitricoxidesynthase ,nNOS)在缺血性老年大鼠海马及皮层神经元中的作用并观察其超微结构变化。方法 建立老年大鼠脑缺血动物模型 ,在不同的时间点进行nNOS免疫组化染色和透射电镜观察 ,检测海马及皮层神经元nNOS的表达并观察其受损情况。结果 缺血 30min后再灌流 12h组nNOS活性显著升高。而假手术组、缺血 30min即刻取材、再灌流 1h ,96h组nNOS几乎无表达 ;6h少量表达 ;2 4h和 48h组中量表达 ;再灌流超过 2 4h组神经元损伤较重。结论 NO在脑缺血发生中对海马及大脑皮层神经元具有毒性作用。 Objective To study the role of nitric oxide synthase and neuronal ultrastructure changes in hippocampus and cerebral cortex neuron after transient cerebral ischemia in the aged rats.Methods The aged SD rat model with cerebral ischemia was set up for studying the expression of nNOS by immunohistochemistry and neuron morphology by transmission electron microscopy (TEM) at different timepoint.Results The activity of nNOS in the 12 h-reperfusing group after 30 minutes of cerebral ischemia was significantly higher than those in other groups. While the group reperfusing over 24 h had a more serious neuron damage.Conclusion NO played an important role in hippocampus and cerebral cortex damage in aged rats with cerebral ischemia.
