摘要
目的 探讨眼球玻璃体内注射CPT cAMP对切断视神经后视网膜节细胞存活的影响。方法 用荧光素逆行示踪标记法和定量解剖学技术观察视神经切断 5、7和 14d后成年金黄地鼠视网膜节细胞的密度。结果 (1)正常视网膜节细胞平均密度为 2 0 0 7± 115 /mm2 ;(2 )视神经切断 5、7、14d后 ,视网膜节细胞平均密度分别下降至 :10 10± 131/mm2 、782± 5 5 /mm2 和2 14± 30 /mm2 ;(3)给予DMSO/生理盐水的对照组在上述各时间段视网膜节细胞平均密度与单纯神经切断组结果相似 ;(4)给予CPT cAMP的实验组在 5、7、14d视网膜节细胞的平均密度分别为 1398± 2 45 /mm2 、12 93± 84/mm2 和 5 0 1± 72 /mm2 ,与视神经切断组和DMSO/生理盐水对照组相比在各时间段上均存在显著性差异 (P <0 0 5 )。结论 CPT cAMP可提高视神经切断后成年金黄地鼠视网膜节细胞的存活。
Objective To investigate the effects of CPT cAMP on survival of axotomized retinal ganglion cell (RGC) by intravitreal injection.Methods Fluorescent retrograde tracing method and quatitative anatomical techniques were used to measure the densities of RGCs in all groups of adult hamsters.Results ①In normal retinas,the mean densities of RGCs were 2007±115/mm 2.②At 5,7,14 days after axotomy,the mean densities of RGCs decreased to 1010±131/mm 2,728±55/mm 2 and 214±30/mm 2,respectively.③In the control group treated with DMSO/sodium saline,the densities of RGCs at 5,7 and 14 days after axotomy were similar to those observed in the axotomized group.④In the group treated with CPT cAMP,the denstities of RGCs at 5,7 and 14 days after axotomy were 1398±245/mm 2,1293±84/mm 2 and 501±72/mm 2,which were different from the axotomized group and DMSO/sodium saline controls(P<0 05).Conclusions CPT cAMP improves RGCs survival after after axotomy in adult hamsters.
