摘要
目的 建立兔再狭窄模型,系统观察球囊血管成形术后血浆P-选择素含量,纤溶酶原激活物抑制剂-1(PAI-1)活性的变化,PAI-1基因表达,以了解PAI-1基因表达与再狭窄间关系。方法 对模型兔进行球囊血管成形术,分别于术前、术后3h,1、2和4周应用ELISA法测定血浆P-选择素含量,应用发色底物法测定血浆PAI-1活性,并利用RT-PCR技术测定不同时期局部血管壁内PAI-1mRNA表达情况。结果 血管成形术后3h,血浆P-选择素含量、PAI-1活性均迅速升高,血管局部PAI-1mRNA在术后3h明显增加达高峰,术后4周达术前水平。结论 血管成形术后,血小板活化,局部PAI-1表达增加,不仅促使损伤局部血栓形成,而且还可刺激局部血管平滑肌增生,并通过抑制局部纤溶活性而促使细胞外基质堆积,在再狭窄形成中起重要作用。
Objective To demonstrate possible association between changes in platelet activity, fibrinolytic potential and restenosis in the balloon - injured rabbit model, following observation on the changes of plasma P - selectin, plasma plasminogen activator inhibitor type 1(PAI - 1 ) activity and the expression of PAI - 1 gene in the arterial wall after vascular injury. Methods In the rabbit model subjected to balloon injury, plasma PAI - 1 activity was determined by chromogenic assay; P - selectin was assayed by ELISA, and PAI - 1 messenger ribonucleic acid (mRNA) expression was assayed with the use of RT - PCR. Results The plasma level of P - selectin increased rapidly from 7.82 ± 0.36ng/ml to 48.43 ± 13.51ng/ml at 3h after balloon injury. PAI - 1 activity in plasma increased sig- nificantly from 7.90 ± 0.87AU/ml to 11.83 ± 1.05AU/ml at 3h after vascular injury. The expression of PAI - 1 mRNA was increased 3.4 - fold compared with that in the control arteries at 3h after vascular injury. Conclusion Our results indicate that balloon injury simulating angioplasty induces high - ac- tivity of the platelet and intramural expression of PAI - 1. The decreased cell surface fibrinolytic activity is likely to result from the increased PAI - 1 expression which may initiate or exacerbate mural thrombosis and contribute to restenosis.
