生长因子网络调节对骨形成作用的研究Ⅰ.TGF-β对人胚成骨样细胞增殖及分化的影响 被引量:17

摘要 在对人胚成骨样细胞分离培养的基础上,进一步探讨不同剂量转化生长因子-β(TGF-β)对人胚成骨样细胞DNA、胶原蛋白和碱性磷酸酶合成的影响。结果表明:TGF-β对体外培养的人胚成骨样细胞DNA合成有抑制作用,对胶原蛋白和碱性磷酸酶合成有促进作用。这两方面的作用在TGF-β0.01~1ng/ml间呈剂量依赖性,1ng/ml时的作用达到最强。本研究提示TGF-β在骨形成中的主要作用可能在于介导机体系统因素对骨细胞的作用。 Osteoblastlike cells were isolated from embryonic human calvarium with modified enzyme digestion method. TGFβ which is believed having biological effects on bone cells was added into the culture medium with seperate concentrations. The incorporation of 3HTdR, 3HProline of osteoblastlike cells and alkaline phosphatase(ALP) content of the cells were tested to study the effects of the TGFβ on the proliferation of osteoblasts.The results showed that TGFβ increased ALP activity and the synthesis of collagen, and the cell proliferation was inhibited. It affected the proliferation and differentiation of osteoblastlike cells with the best 1 ng/ml among four concertration of 0.01 ̄10 ng/ml. These findings suggested that function of TGFβ might be modified by other local growth factors.
