摘要
在对人胚成骨样细胞分离培养的基础上,进一步探讨不同剂量转化生长因子-β(TGF-β)对人胚成骨样细胞DNA、胶原蛋白和碱性磷酸酶合成的影响。结果表明:TGF-β对体外培养的人胚成骨样细胞DNA合成有抑制作用,对胶原蛋白和碱性磷酸酶合成有促进作用。这两方面的作用在TGF-β0.01~1ng/ml间呈剂量依赖性,1ng/ml时的作用达到最强。本研究提示TGF-β在骨形成中的主要作用可能在于介导机体系统因素对骨细胞的作用。
Osteoblastlike cells were isolated from embryonic human calvarium with modified enzyme digestion method. TGFβ which is believed having biological effects on bone cells was added into the culture medium with seperate concentrations. The incorporation of 3HTdR, 3HProline of osteoblastlike cells and alkaline phosphatase(ALP) content of the cells were tested to study the effects of the TGFβ on the proliferation of osteoblasts.The results showed that TGFβ increased ALP activity and the synthesis of collagen, and the cell proliferation was inhibited. It affected the proliferation and differentiation of osteoblastlike cells with the best 1 ng/ml among four concertration of 0.01 ̄10 ng/ml. These findings suggested that function of TGFβ might be modified by other local growth factors.
