摘要
目的 :研究紫外线辐射对小鼠血清肿瘤坏死因子 α(TNFα)水平和脾细胞 TNFα m RNA表达的影响 ,从而探讨紫外线辐射所致免疫抑制的作用机制。方法 :用酶联免疫吸附试验检测小鼠血清中 TNFα含量。一步法提取脾细胞 RNA,逆转录聚合酶链反应检测其 TNFα m RNA表达。结果 :一定剂量的紫外线辐射不能使小鼠血清中出现可检测的 TNFα(P>0 .0 5 )。但可使诱生的 TNFα水平下降 (P<0 .0 1) ,辐射剂量与 TNFα的浓度呈直线负相关 (P<0 .0 1)。脾细胞 TNFα m RNA表达被抑制 (P<0 .0 0 1)。结论 :紫外线辐射可能是通过抑制具有分泌 TNFα功能的淋巴细胞和 (或 )单核巨噬细胞的 TNFα m RNA转录 ,进而抑制 TNFα的分泌 。
Objective:To investigate the mechanisms of immunosuppression induced by ultraviolet radiation, and observed the influence of ultraviolet radiation on serum TNFα level and spleen cells TNFα mRNA expression in mice. Methods:Serum TNFα was detected by Enzyme Linked Immuno Sorbent Assay (ELISA) method. A step extraction method was used to get RNA of spleen cells. mRNA expression of TNFα was assayed by RT PCR test. Results: Ultraviolet radiation failed to induce a detectable rise in circulating TNFα in mice( P>0.05 ). TNFα secretion was significantly inhibited by ultraviolet radiation ( P<0.01) . The dosage of ultraviolet radiation was negatively correlated with the serum TNFα level ( P<0.01) , and the mRNA expression of TNFα in spleen cells was suppressed by ultraviolet radiation ( P<0.001) . Conclusion:The results suggest that ultraviolet radiation may suppress TNFα mRNA transcription in macrophage or activate lymphocytes further to diminish its TNFα secretion and finally influence some immune responses.
