摘要
目的 :探讨诱导型一氧化氮合酶 (iNOS)在系膜增殖性肾小球肾炎 (MsPGN )中的表达及意义。方法 :采用逆转录 -聚合酶链反应方法检测 36例系膜增殖性肾炎 (MsPGN)、18例非MsPGN患者肾活检组织及 16例正常肾组织中诱导型一氧化氮合酶 (iNOS)mRNA的相对表达水平。结果 :MsPGN组、非MsPGN组iNOSmRNA表达分别为 :1 0 2 9± 0 .16 6、0 .4 59± 0 .115,MsPGN组明显高于非MsPGN组 (P <0 .0 0 1)。中度MsPGN组 (1 0 92± 0 139)明显高于轻度MsPGN组 (0 .94 9± 0 .16 7) (P <0 .0 1)。正常肾组织中无iNOSmRNA表达。尿蛋白 >3.5g/d组与 <3.5g/d组iNOSmRNA表达分别为 :1.0 98± 0 .137、0 .92 2± 0 .156 ,二者有显著性差异 (P <0 .0 1)。结论 :iNOS在MsPGN的发病和进展机制中起重要作用。
Objective:To investigate expression of inducible NO synthase(iNOS) mRNA and its significance in the kidneys patients with mesangial proliferative glomerulo nephritis(MsPGN).Methods:Biopsied renal tissues from 36 patients with MsPGN,18 patients with non MsPGN and 16 patients with normal renal tissues were included in this study. iNOS mRNA was detected by semi quantitative reverse transcription polymerase chain reaction.Results:The iNOS mRNA level in the patients with MsPGN and non MsPGN were (1.029±0.166) and (0.459±0.115) respectively.There were significant differences between the two groups (P<0.001).The level of iNOS mRNA in MsPGN patients with medium mesangial proliferation(1.092±0.139) was much higher than that in the patients with light proliferation(0.949±0.167)(P<0.01).Normal renal tissues did not show any iNOS mRNA expression.The level of iNOS mRNA expression in patients with proteinuria>3.5g/d( 1.098 ±0.137) was higher than that in patients with proteinuria<3.5g/d(0.922±0.156)(P<0.01).Conclusion: iNOS mRNA expression plays an important role in the pathogenesis of MsPGN.
