摘要用继代6年的八倍体小黑麦单倍性胚性愈伤组织,建立悬浮系。取悬浮细胞分离原生质体,培养在附加2,4-D 2mg/L、KT 0.2mg/L、NAA 0.1mg/L、琼脂糖0.5%的 MS 培养基中。培养5天后见到第一次分裂,3周后发育成肉眼可见的小愈伤组织。此时添加降低糖浓度的新鲜培养基。再生愈伤转移到附加2,4-D 1mg/L 的 MS 固体培养基上后生长迅速。在无任何激素的 MS 培养基上分化,得到再生绿苗。
