摘要
目的 T3对细胞的代谢调节作用是通过甲状腺激素核受体 (T3R)介导的 ,细胞中T3R量与细胞对T3的反应性有关。本文探讨了T3R表达与人体不同甲功状态的关系。方法 应用半定量RT PCR法 ,研究了Graves 甲亢、桥本甲炎甲功正常、桥本甲炎甲减患者外周血白细胞中T3R各亚型mRNA表达量的变化 ,并与正常对照进行比较。结果 外周血白细胞中T3Rα1 、T3Rα2 、T3Rβ2 mRNA表达在Graves 甲亢患者降低 ,在桥本甲炎甲减患者升高 ,而桥本甲炎甲功正常者有升高倾向 ;T3Rβ1 mRNA在不同甲功状态中变化不大。结论 在外周血白细胞中 ,T3RmRNA各亚型表达与甲功状态密切相关 ,T3对其受体亚型mRNA表达水平有下调作用 ,但T3Rβ1 mRNA表达不受甲功状态影响。
Objective The metabolic effects of T 3 are mediated by binding to its nuclear receptors(T 3Rs). The relationship between the T 3Rs mRNA expression and different thyroid states was discussed. Methods By using Semiquantitative RT-PCR method, the researchers examined T 3R mRNAs level in peripheral leukocytes of Graves disease (GD), Hashimoto thyroiditis(TH) with euthyroidism, Hashimoto thyroiditis with hypothyroidism and normal controls. Results T 3Rα 1, T 3Rα 2, T 3Rβ 2 mRNA levels were all decreased in GD patients, while increased in HT with hypothyroidism. Conclusion They all was tend to increase in HT with euthyroidism. T 3Rβ 1 mRNA level was no major difference in each group.
