摘要
目的 探讨胆汁中可溶性细胞间粘附分子 - 1(s ICAM- 1)表达水平与大鼠肝移植急性排斥反应的关系。方法 将大鼠分为单纯胆汁外引流组、同种同基因肝移植组和同种异基因肝移植组 ,后二组行原位肝移植加胆汁外引流 ,用 EL ISA法检测未移植时及移植后 2、4、7d胆汁中 s ICAM- 1表达水平。结果 同种异基因肝移植组大鼠胆汁中 s ICAM- 1表达水平显著高于同种同基因肝移植组 ,且其水平升高与移植术后时间变化呈正相关。结论 检测移植肝组织中 s ICAM- 1的表达水平升高 ,有助于肝移植急性排斥反应的诊断 ,并可动态监测其严重程度。
Objective To investigate the relationship between the expression of soluable intercellular adhesion molecule 1(sICAM 1) and acute liver rejection by studying the expression feature of the molecule in bile.Methods Forth eight SD rats and seventeen Wistar rats were divided into three experiment groups randomly.Group A underwent biliary extra drainage only.Group B,C underwent orthotopic liver transplantation with biliary extra drainage.Group B served as isograft group,both donors and recipients were SD rats.Group C served as allograft group,wistar rats were used as donors and SD rats were served as recipients.All the recipients were examined on days 2、4 and 7 following orthotopic liver transplantation respectively.Paraff inembidded,and HE stained sections were examined microscopically to classify acute rejection degree according to the Kemnitz system.The titer of sICAM 1 in bile was tested by enzyme linked immunoabsorbent assay method.Results In allograft group,the level of sICAM 1 was markedly higher than that in either isograft group or normal group,and the expression level of the molecule was in proportional to the postoperative time.Conclusion The upregulation of sICAM 1 in bile is helpful to the diagosis of acute rejection after liver transplantation,and positively relative to the degree of acute liver rejection.
