摘要
目的:了解长春新碱(VCR)诱导的自噬性细胞凋亡是否与线粒体跨膜电位(△Ψm)改变有关及其可能的机制。材料与方法:以正常人的肝细胞系为体外模型,应用碘化丙啶(PI)/Rhodamine123(Rh123)双重染色检测△Ψm,通过亚G1期细胞和透射电镜鉴定细胞凋亡。结果与结论:VCR可明显诱导线粒体△Ψm下降,也可以诱导L-02细胞自噬性凋亡。线粒体△Ψm下降可能是VCR诱导自噬性细胞凋亡的关键环节。
Purpose: To understand if vincristine (VCR) is related to the alteration of mitochondrial transmembrane potentials (A) and its possible mechanisms. Methods: Human liver cell line L-02 was used as in vitro models. The was detected by the double staining of propidium iodide (PI) and Rhodamine 123(Rhl23),which fluorescent intensities were measured by flow cytometry, while apoptosis was confirmed by sub-G1 cell content as well as transmission electron microscopy observation. Results and Conclusion: In the cells sensitive to VCR-induced autophagic apoptosis, the was disrupted with VCR treatment. Vincristine can induced L-02 cell autophagic apoptosis, which could associate closely with the collapse. The disruption of the maybe is the key event of VCR-induced autophagic apoptosis.
