以抗坏血酸-Fe^(3+)作为自由基生成体系,结合琼脂糖电泳和激光密度扫描等方法,观察了自由基对质粒pBR322 DNA的损伤及过氧化氢酶和超氧化物歧化酶的防护效能。结果表明,该体系生成的羟自由基可造成DNA的单链断裂而使pBR322的超螺旋构型转变为开环构型;过氧化氢酶对羟自由基所致DNA损伤的抑制作用显著,而超氧化物歧化酶无明显抑制作用。
Exposure of the plasmid pBR322 to hydroxyl radicals generated from ascorbate/iron system introduced single strand scissions and conformational modifications. The latter were observed by means of agarose electrophoresis and were quantitatively analyzed by laser density-scanning. The DNA damage induced by hydroxyl radicals was completely prevented by catalase, whereas superoxide dismutase was much less effective.
