高压氧中毒小鼠大脑皮质毛细血管ALP活性的变化

CHANGES OF ALP ACTIVITY IN THE MICE CEREBRAL CORTEX CAPILLARY AFTER HYPERBARIC OXYCEN POISONING

摘要 采用电镜细胞化学和酶活性的图象定量分析方法,对高压下氧中毒小鼠大脑皮质毛细血管内皮细胞的碱性磷酸酶(ALP)活性定位和强度变化进行了研究。许多定量参数表明,高压下发生氧中毒后,小鼠大脑皮质毛细血管内皮细胞ALP活性增强。作者讨论了毛细血管ALP活性增高与功能改变之间的可能关系。 he Changes of alkaline phosphatase(ALP) activity in the mice cerebral cortex capillary endotheli-um after hyperbaric oxygen poisoning(HOP)were studied using the methods of ultra-cytochemistry and image quantitative analysis,15 ICR mice weighing approxiniately 20~22 g were randomly divided into 3 groups:the normal (NP) group, the hyperbaric air(HA)group,and the hyperbaric oxygen poisoning group(HOP).The animals of HA and HOP groups were put into the same hyperbaric aninial chamber and were given air and 100%oxygen respectively.The pressure was increased from 0.1 MPa(1 ATA)to 0.5 MPa(5 ATA)in 4 min and the animals were kept at 0.5MPa for 37 min. The time of decom-pression was 15 min. The cerbral cortex of all the mice were then removed and fixed according to the method of ultra-cytochemistry. Quantitative parameters of ALP activity in the cerebral cortex capillary endothelium were measured and calculated by means of a computer image analyser.The results showed that there were no significant change of structure and ALP activity of capillary endothelium in the HA group; whereas in the HOP group,some mitochondria in some neurons and cap-illary endothelium showed vacuolation with broken cristae and the capillary endothelial membrane was injured;at the same time,the ALP activity in the membrane increased significantly. The ALP is an unspecialized enzyme which hydrolyses the phosphate. It is mainly located at the cell membrane and takes part in the transport process of cells. The increase of ALP activity in the HOP group might mean a compensative or pathological increase of transporting function. Under this circum-stance,the blood-brain barrier in the HOP group could be jeopardized
