摘要
目的:观察大黄对急性肝衰竭大鼠细胞凋亡及Bcl-2和Caspase-3表达的影响,旨在探讨其防治急性肝衰竭的作用机制。方法:30只Wistar大鼠随机分为正常组、模型组和大黄组。大黄组大鼠造模前3 d予大黄水煎液10 g/(kg·d)灌胃,采用D-氨基半乳糖(D-Ga1N)/内毒素脂多糖(LPS)腹腔注射造成大鼠急性肝衰竭模型。造模6 h后,取血检测血清谷草转氨酶(AST)、谷丙转氨酶(ALT)和总胆红素(TBIL)含量,并开腹取大鼠肝组织,应用原位末端标记(TUNEL)法检测肝细胞凋亡情况,应用免疫组化法分别检测肝组织中Bcl-2和Caspase-3蛋白表达情况。结果:与正常组比较,模型组大鼠血清AST、ALT、TBIL含量和肝细胞凋亡指数(AI)及Caspase-3蛋白表达均明显升高(均P<0.01),Bcl-2蛋白表达明显下降(P<0.01);与模型组比较,大黄组大鼠血清AST、ALT和TBIL含量、AI及Caspase-3蛋白表达均显著下降(均P<0.01),而Bcl-2蛋白表达明显上调(P<0.01)。结论:大黄可明显减轻急性肝衰竭大鼠肝脏损伤,其作用机制可能与上调Bcl-2表达、下调Caspase-3表达,从而抑制肝细胞凋亡有关。
Objective:To observe the effects of of rheum officinale on hepatocyte apoptosis and expressions of Bcl-2 and Caspase-3 in rats with acute liver failure and to investigate the possible mechanism. Methods : Thirty Wistar rats were divided into normal group, model group and rheum officinale group. The rheum officinale group was given rheum officinale solution 10 g/ ( kg · d ), and 3 days consecutively. 1 hour after the last time, the acute liver failure model was reproduced in rats by peritoneal injection of D-GalN and LPS. After six hours, the liver tissue of rats was taken out for TUNEL assay of hepatocyte apoptosis, and Bcl-2 and Caspase-3 protein expressions and detection of liver tissue were detected by immunohistochemistry. Results : Compared with the normal group, Bcl-2 protein expression was significantly decreased in the liver tissue of the model group and the liver apoptotic index and Caspase-3 protein expression were significantly increased (P〈0.01). Compared with the model group, hepatocyte apoptosis index and Caspase-3 protein expression in the group of rheum officinale were significantly downregulated, while Bcl-2 protein expression was significantly up-regulated ( P〈0.01 ). Conclusion : Rheum officinale has protective effect on the apoptosis of rats with acute liver failure and its mechanism may be related to up-regulation of Bcl-2 expression, down-regulation of Caspase-3 expression and thus the apoptosis of hepatocyte is repressed.
