摘要
本试验通过PCR方法对17株广西猪源猪链球菌2型(SS2)荚膜多糖(CPS)基因cps2J片段进行了扩增、克隆测序,同源性分析结果表明,17株广西SS2的cps2J基因核苷酸和氨基酸的同源性分别在91.9%~99.8%和89.1%~99.7%;菌株可以分为Ⅰ群和Ⅱ群,包括5株发病猪源菌株的16株菌均分在Ⅰ群,Ⅱ群只有GXWX193株。Ⅰ群菌株的CPS基因核苷酸和氨基酸的同源性分别在96.8%~99.8%和94.6%~99.7%,与参考菌(荷兰Netherland、猪源菌株江苏ZYS8和四川SC17、人源菌株江苏98015)的核苷酸和氨基酸的同源性分别在96.4%~99.9%和94.1%~100%。Ⅱ群菌株与参考菌株的核苷酸和氨基酸同源性分别在91.3%~92.4%和89.1%~91.0%。17株广西猪源SS2菌株的cps2J基因序列变异程度小。
Amplified and cloned by PCR, then sequenced the virulence-associated factor cps2J gene of 17 strains of type 2 Streptococcus suis isolated from Guangxi. The homologies of nucleo-tide and predicted amino acids of 17 Guangxi SS2 are 91.9%~99.8% and 89.1%~99.7% respectively.17 SS2 strains could be divided into two groups as Ⅰ (11 strains isolated from healthy pigs and 5 strains from diseased pigs) and Ⅱ (1 strain isolated from healthy pigs). The homologies of group I were very high (96.8%~99.8% nt and 94.6%~99.7% aa). Compared to reference strains(Netherland/ZYS8/SC17/98015), homologies of group Ⅰ were 96.4%~99.9%nt and 94.1%~100%aa , 91.3%~92.4%nt and 89.1%~91.0%aa in group Ⅱ. There is a low degree of variation in cps2J gene in 17 SS2 isolated from pigs in Guangxi ,China.
