摘要
目的:综合评价DRD2基因TaqIA多态性与兴奋型毒品依赖(可卡因、甲基苯丙胺)遗传易感性的相关性。方法:按照统一的检索策略,检索PubMed、Medline、Web of Science、中国学术期刊全文数据库(CNKI)、万方数据库和VIP数据库有关DRD2基因TaqIA多态性与兴奋型毒品依赖遗传易感性相关研究,截止时间2013年10月。按纳入和排除标准纳入合格文献,提取数据并进行质量评价,用Stata 11.0软件在同质、异质、显性和隐性遗传模式下进行meta分析。结果:共纳入9篇文献(5篇研究可卡因依赖,4篇研究甲基苯丙胺依赖),收集病例2 144例和对照2 245例。Meta分析结果显示DRD2基因TaqIA多态性与兴奋型毒品依赖遗传易感性在4种遗传模式下均无统计学意义(P>0.05),按毒品依赖种类、种族和质量评分进行亚组分析亦没有发现显著的统计学意义(P>0.05)。DRD2基因TaqIA多态性与兴奋型毒品依赖遗传易感性在同质和显性遗传模式下存在显著异质性,Noble EP、Ballon N和Han DH文献可能是异质性来源。结论:DRD2基因TaqIA多态性可能与兴奋型毒品依赖(可卡因、甲基苯丙胺)遗传易感性没有相关性。
Objective:Synthetically estimate the association between DRD2 gene TaqlA polymorphism and stimulant drug( cocaine, methamphetamine) dependent susceptibility. Methods:According to the uniform retrieval strategy,search the information about DRD2 gene TaqIA polymorphism and stimulant drug (cocaine, methamphetamine) dependent susceptibility from the electronic database PubMed, Medline, Web of Science, CNKI,WanFang and VIP up to Oct,2013. The quality of the included studies was evaluated and the data was extracted. The pooled OR and 95% CI, and the heterogenicity test were calculated under different genetic models by Stata 11.0 software, including heterozygote ( TC vs CC ) , homozygote ( TT vs CC ) , dominant ( TC/TT vs CC) , and recessive ( TT vs CC/TC ) genetic models. Results:A total of 9 case-control studies were identified(5 studies in cocaine,4 in methamphetamine) ,involving 2 144 cases and 2 245 controls. The current meta-analysis showed that the DRD2 Taql A polymorphism was no significantly associated with stimulant drug( cocaine, methamphetamine) dependent susceptibility under all genetic model(P 〉 0.05 ). Subgroup analysis results were consist- ent with the main analyses by drug type, ethnicity and quality score( P 〉 0. 05 ). The PQ values and I2 showed existed heterogeneity under homozygote and dominant comparison models. We found that Noble EP,Ballon N and Han DH might be the sources of heterogeneity. Conclusion:The current recta-analysis suggested that DRD2 TaqlA polymorphism might not be associated with stimulant drug( eoeaine,methamphetamine) dependent susceptibility.
