固定化α-淀粉酶的制备及性能研究

摘要 以P(DMAA-co-AM)凝胶为载体,制备了固定化α-淀粉酶,并对其性能进行了研究.研究结果表明:α-淀粉酶被载体固定化后活性受温度和pH的影响降低;固定化酶重复操作6次后,酶活仍可达到73%;在4℃环境中储存70天,酶活性由固定前的26%提高到约60%;固定化酶和游离酶的米氏常数Km分别为1.33,5.16mg/mL,最大反应速率Vmax分别为0.52,2.56mg/min. The immobilizeα-amylase was prepared by taking P(DMAA-co-AM)hydrogel as carrier.The results showed that the effect of temperature and pH on the activity ofα-amylase dropped,and the activity of immobilized a-amylase remained 73% higher at sixth-turn.The activity of immobilizationα-amylase increased from 26%to about 60% under the temperature of 4℃ and storage for 70days.Kinetic studies demonstrated that Kmvalues were 1.33mg/mL and 5.16mg/mL for immobilized and freeα-amylase respectively,and Vmaxvalues were 0.52mg/min and 2.56mg/min for immobilization and freeα-amylase respectively,too.
