摘要
本试验旨在研究博落回中血根碱、白屈菜红碱、原阿片碱和别隐品碱在不同浓度、不同时间段对猪肠上皮细胞(IPEC-1)增殖的影响。IPEC-1预培养24 h后,在培养基中分别加入不同浓度(4、2、1、0.5、0.25、0.1、0.05、0.025、0.012 5、0.006 25 μg/mL的血根碱或白屈菜红碱;100、50、25、12.5、6.25、3.125、1.562 5 μg/mL的原阿片碱或别隐品碱,各组不同浓度梯度均设8个重复)的博落回生物碱继续培养12或24 h,4-甲基偶氮唑盐(MTT)染色法检测细胞增殖。结果表明:0.25~4 μg/mL的血根碱、0.25~4 μg/mL的白屈菜红碱、100 μg/mL的原阿片碱或100 μg/mL的别隐品碱对IPEC-1的增殖有显著的抑制作用(P0.05)。由此可见,博落回生物碱在一定浓度范围内可促进IPEC-1的增殖。这说明博落回生物碱提高动物生长性能可能是通过促进小肠细胞增殖分化更新,从而增强小肠消化、吸收、免疫屏障和应激反应等功能的途径实现的。
To study the effects of sanguinarine, chelerythrine, protopine and allocryptopine from Macleaya cordata on intestinal porcine epithelial cells (IPEC-1) proliferation at different concentrations and different periods. IPEC-1 cells were pre-incubated for 24 hours, and then exposed to different concentrations (4, 2, 1, 0.5, 0.25, 0.1, 0.05, 0.025, 0.012 5 and 0.006 25 μg/mL of sanguinarine or chelerythrine, and 100, 50, 25, 12.5, 6.25, 3.125 and 1.562 5 μg/mL of protopine or allocryptopine; each treatment had 8 replicates) for additional 12 or 24 hours, respectively, cell viability was measured by MTT assay. The results showed as follows: after the IPEC-1 cells were treated by 0.25 to 4 μg/mL of sanguinarine, 0.25 to 4 μg/mL of chelerythrine, 100 μg/mL of protopine or 100 μg/mL of allocryptopine for 12 hours, the proliferation of cells was significantly inhibited (P0.05). The results indicate that the Macleaya cordata alkaloids can promote the proliferation of IPEC-1 under certain concentrations; it speculates that Macleaya cordata alkaloids may improve the intestinal digestive, absorptive, immune and barrier function through this way, and then improve the animal growth performance.
