摘要
类胰岛素生长因子(Insulin-like grow factor,IGFs)与动物生长发育呈强正相关,为了填补鸭胚胎期类胰岛素生长因子受体(Insulin-like growth factor-I receptors,IGF-IR)在骨骼肌发育变化的研究空白,实验采用qRT-PCR研究生长速度差异较大的高邮鸭和金定鸭胚胎期(13、15、17、19、21和23胚龄)骨骼肌(胸肌和腿肌)成肌细胞IGF-IR基因发育性表达的差异。研究显示,成肌细胞IGF-IR mRNA在2个品种的各发育时间都表达,种间同一组织表达量变化趋势较一致,总体均呈现先升后降的趋势。胸肌中,基因表达量在17胚龄时达到最高峰,显著高于其他胚龄(P<0.05);腿肌中13胚龄与17胚龄都显著高于其他胚龄(P<0.05);品种间除了金定鸭17胚龄时胸肌IGF-IR mRNA的表达量极显著高于高邮鸭(P<0.01),其他各胚龄差异均不显著。不同肌肉组织的IGF-IR mRNA表达变化存在种间和空间差异性。不同肌肉组织成肌细胞中的IGF-IR基因表达具有显著差异,与对应组织中的表达不一致,揭示在研究转录分析时应从不同层次进行比较研究,以期获得比较系统的科学支撑。
Insulin-like growth factor has a strong positive correlation with animal growth and development . In order to fill the gap in the research on the changes of insulin-like growth factor-I receptors ( IGF-IR) dur-ing the development of the skeletal muscle of duck , the IGF-IR mRNA expressions on the skeletal myoblast during the embryonic stage of Gaoyou duck and Jinding duck i .e.on day 13,15,17,19,21 and 23 were detec-ted using quantitative RT-PCR method in this experiment .The result indicated that the profiles of duck IGF-IR gene on myoblast IGF-IR mRNA in the two duck breeds were relatively consistent during the early develop-ment.On the whole the expression level of IGF-IR gene increased first and decreased afterwards and showed an analogous curves between the varieties .The highest level of IGF-IR mRNA appeared at the 17 embryo age in pectorals and at the 13 and 17 embryo ages in leg of both the breeds ( P〈0.05) .The expression levels of IGF-IR mRNA in pectorals of Jinding duck were significantly higher than that of Gaoyou duck at the 17 embryo age ( P〈0.01) and others had no significant difference ( P〉0.05) between varieties.Conclusion:there were differ-ences between varieties and spaces in the expression of IGF-IR mRNA in different muscle myoblast .There were significant difference between different skeletal myoblasts and inconsistent with the corresponding tissues in the expression level of IGF-IR gene.So the transcriptional analysis of gene expression should focus on differ-ent layers in order to obtain a more systematic scientific support .
