摘要
目的 :建立反相高效液相色谱测定西红花甙提取物中西红花甙 1和西红花甙 2含量的方法。方法 :采用No va PakC18(4μm ,15 0mm× 3 .9mm)色谱柱 ,流动相甲醇 水 (5 5∶45 ) ,流速 1ml·min-1,检测波长 440nm。结果 :西红花甙 1对照品的线性范围为 0 .0 3~ 0 .3 0g·L-1,r=0 .9999,平均加样回收率为 10 0 .2 1% ,RSD =0 .3 8% ;西红花甙 2对照品的线性范围为 0 .0 5~ 0 .3 0g·L-1,r =0 .9999,平均加样回收率为 99.78% ,RSD =0 .42 %。结论 :采用反相高效液相色谱法测定西红花提取物中西红花甙 1和西红花甙 2含量 ,操作简便。
Objective To apply RP HPLC method to determine the content of crocusⅠ and crocusⅡ in crocus sativus L.Methods The HPLC instrument was equipped with Nova Pak C 18 (4?μm,150?mm×3.9?mm) column. The mobile phase was methanol water(55∶45).The flow rate was 1?ml·min -1 .The UV detector wavelength was set at 440?nm.Results The standard curve of crocusⅠwas linear in the concentration range of 0.03~0.30?g·L -1 ( r =0.999?9),the average recovery rate was 100.21% and relative standard was 0.38%( n =5).The standard curve of crocus Ⅱ was linear in the concentration range of 0.05~0.30 g·L -1 ( r =0.999?9),the average recovery rate was 99.78% and relative standard was 0.42% ( n =5).Conclusion The method is simple, sensitive,and accurate to determine the content of crocusⅠand crocus Ⅱ in crocus L.
