摘要
目的:探讨温阳化瘀方对糖尿病周围神经病变(DPN)模型大鼠炎症因子及其相关因子的调节作用。方法:大鼠随机分为正常对照组、高脂高糖(模型)组、弥可保组、二甲双胍(降糖)组、温阳化瘀方高(18g/kg)、中(12g/kg)、低(6g/kg)剂量组。除正常对照组外,各组大鼠禁食12h,按60 mg/kg剂量尾静脉一次性快速注射链佐菌素制备动物模型,于模型制作当日各给药组大鼠分别ig给予药物治疗,每日一次,连续12周,其中降脂组给予弥可保1.25 mg/kg,降糖组给予二甲双胍670 mg/kg,试药高、中、低剂量组分别给予温阳化瘀方浓缩液18g/kg、12g/kg、6g/kg,正常对照组、模型组给予同样量的生理盐水。给药最后一天取血测定空腹血糖;取血清采用酶联免疫吸附分析方法测定血清胰岛素(INS)、瘦素(LEP)、白细胞介素(IL)-6、C-反应蛋白(CRP)、肿瘤坏死因子-α(TNF-α)、神经生长因子(NGF)、胰岛素生长因子(IGF-1)水平,计算胰岛素敏感指数。结果:温阳化瘀方18g/kg、12g/kg、6g/kg剂量组均能显著降低DPN模型大鼠空腹血糖有降低作用;18g/kg、12g/kg、6g/kg剂量组均能显著升高DPN模型大鼠INS、LEP水平,调节ISI,能使IL-6、CRP、TNF-α等炎症因子调节到正常水平,与模型组比较,具有非常显著的差异。本方对DPN大鼠血清神经生长因子(NGF)、胰岛素生长因子(IGF)-1有一定调节作用,与模型组比较,18g/kg、12g/kg、6g/kg三个剂量组均显示有非常显著的差异。研究结果表明,温阳化瘀方最小有效剂量为6g/kg,其有效剂量范围是6g^18g/kg。结论:温阳化瘀方对DPN模型大鼠INS、炎症因子均有一定调节作用,提示温阳化瘀方有调节脂肪和糖代谢的作用,对慢性糖尿病所致周围神经病变有一定治疗作用。
Objective: To investigate effects of the Wenyanghuayu decoction on inflammation factors and relative factors of in diabetic peripheral neuropathy( DPN) model rat. Method: Take the male SPF level rat,the random divides normal control group,high fat and sugar( model) forms; high fat and sugar + methylcobalamin,high fat and sugar + treated with metformin; high fat and sugar treated with Wenyanghuayu decoction large dose,middle dose and small dose groups. The rats of every group were taken in high fat and high sugar and taken in addition to the normal control group,a group rats fasting 12 h,intravenous single rapid injection preparation of animal model of streptozotocin 60 mg / kg by except control group. In addition to the normal control group,other groups of rats were feeding with high fat and high sugar. In addition to the normal control group,other groups and model at all for set of rats given by ig drug treatment,once a day,for 12 weeks. Each 10 d weighs every group animal weight once. Among them,the rats in lipid-lowering group were taken 1. 25 mg / kg methycocalamin; hypoglycemic group rats 670 mg / kg metformin; Wenyanghuayu decoction large dose group,middle dose group and small dose group rats 18 mg / kg,12 mg / kg and mg / kg Wenyanghuayu decoction concentrated solution differently. Control group and model group rats were taken normal salt administration by gavage by equal content. The level of serum insulin,interleukin( IL)-6,C-reactive protein( CRP),tumour necrosis factor- alpha( TNF-α),nerve growth factors( NGF) and insulin growth factors( IGF)-1 level were detected by enzyme-linked immunosorbent assay( ELISA). And insulin resistance( HOMA-IR) index was counted. Results: The level of insulin was of a very significant difference after the decoction was taken for 12 weeks while being compared with control group and model group( all P < 0. 01).
