复脉膏外敷对大鼠长春瑞滨外渗性损伤的影响 被引量:5

Study on effects of Fumai paste on pathology of skin of rat experiment model of vinorelbine-induced extravasation injury,and effects of VEGF and EGFR of tissue

摘要 目的:通过复脉膏外敷对大鼠长春瑞滨外渗性损伤组织的病理观察,并研究其对外渗性损伤组织中VEGF、EGFR的影响,以探讨复脉膏外敷防治化疗性静脉炎的疗效及机制。方法:42只大鼠随机分为两组:复脉膏组、50%硫酸镁组,两组大鼠左侧损伤处分别予复脉膏外敷、50%硫酸镁湿敷;右侧予生理盐水外敷作为对照组。在损伤后第1、4、7、11、14、18天,分别随机取5只大鼠,测量外渗性损伤部位;并分别随机取3只大鼠,切除损伤部位的皮肤及皮下组织,以备观察。观察指标:皮损面积、病理观察、VEGF及EGFR的表达。结果:复脉膏(0.2g)外敷较硫酸镁湿敷及生理盐水外涂能明显减小各时间点的平均皮损面积。复脉膏组损伤皮肤早期炎症反应较硫酸镁组及对照组为轻,未见出血及血栓形成,后期可见损伤部位有充足的肉芽组织及新鳞状上皮生成。复脉膏(0.2g)外敷较硫酸镁湿敷及生理盐水外涂能明显刺激损伤组织中VEGF的阳性表达。复脉膏(0.2g)外敷较硫酸镁湿敷及生理盐水外涂能更早更强刺激损伤皮肤中EGFR的阳性表达。结论:复脉膏外敷于损伤皮肤后能上调VEGF的表达,促进EGFR的快速增加,进而促进损伤皮肤组织的愈合。 Objective: By studying on effects of Fumai Paste on pathology of skin of rat experiment model of vinorelbine-induced extravasation injury,and effects of VEGF and EGFR of tissue; then discuss the curative effect and mechanism of treatment of chemotherapeutic phlebitis with external application of Fumai paste. Methods: Forty-two rats were divided randomly into two groups with 21 rats in each group. Each rat was established experimental model. Each rat of group of Fumai paste was treated with external application of Fumai paste on left side; each rat of group of magnesium sulfate was treated with hydropathic compress of 50% magnesium sulfate on left side. The right side of each rat were control group,and were treated with hydropathic compress of normal saline. Five rats were selected randomly from each group in 1day,4days,7days,11days,14days and 18days,and the area of extravasation injury of each rat was measured. Three rats were selected randomly from each group,and the skin and subcutaneous tissue of extravasation injury of each rat was cut off. The obvervational index include: the area of extravasation injury、pathology of skin、expressions of VEGF and EGFR of tissue. Results: Fumai paste( 0. 2g) could reduce the average area of extravasation injury of each point-in-time more than 50% magnesium sulfate and normal saline( P < 0. 01). In early time,the inflammatory response of group of Fumai paste was lighter than which of group of magnesium sulfate and control group. There was no bleeding and thrombogenesis of group of Fumai paste. In late time,the skin and subcutaneous tissue of extravasation injury of group of Fumai paste generated adequate granulation tissue and new squamous epithelium. Fumai paste( 0. 2g) could stimulate the positive expression of VEGF of tissue of animal experiment model of vinorelbine-induced extravasation injury more than 50% magnesium sulfate and normal saline( P < 0. 01).
