摘要
目的:观察当归补血汤含药血清对血虚模型小鼠骨髓造血功能的影响,阐述当归补血汤补气生血作用机理。方法:连续4天腹腔注射环磷酰胺30mg/kg复制化学性血虚小鼠模型;灌胃给予15g/kg、7.5g/kg、3.75g/kg当归补血汤水煎液,一日两次,第4天腹主动脉取血,分离含药血清;将各剂量当归补血汤含药血清加入到血虚模型小鼠骨髓细胞的培养体系中,观察各组含药血清对骨髓造血祖细胞CFU-GM、CFU-E、CFU-B及骨髓有核细胞DNA合成的影响。结果:当归补血汤15g/kg含药血清5%组、10%组,当归补血汤7.5g/kg含药血清10%组、15%组,以及当归补血汤3.75g/kg含药血清15%组对血虚小鼠骨髓CFU-GM的集落生成均具有促进作用;当归补血汤15g/kg含药血清10%组、20%组,当归补血汤7.5g/kg含药血清20%组、30%组可明显升高骨髓CFUE和BFU-E集落个数;当归补血汤15g/kg含药血清0.5%组,7.5g/kg含药血清1%组、2%组及3.75g/kg含药血清1%组、2%组对血虚小鼠骨髓有核细胞DNA的合成具有促进作用。结论:当归补血汤含药血清能够促进骨髓粒系、红系造血祖细胞的增殖与分化,促进骨髓有核细胞DNA的增殖,达到其补气生血的功效。
Objective: The effection of Danggui Buxue Decoction serum on blood deficiency mice bone marrow hematopoietic function was observed to explain the mechanism of Danggui Buxue Decoction for Supplementing Qi and activating blood circulation. Methods: The bone marrow nucleated cells of blood-deficient mice were cultured in vitro,and different doses of Danggui Buxue Decoction serum were used to observe the effection on CFU-GM,BFU-E and DNA content. Result: The number of CFU-GM in Danggui Buxue Decoction serum high doses of 5%group,10% group,middle dose of 10% group,and low doses of 15% group,the number of CFU-E colony in high doses of 10% group,20% group,and middle dose of 20% group,30% group,the number of BFU-E colony in middle dose of 20% group,30% group were increased significantly. Bone marrow nucleated cells DNA in high doses of 0. 5% group,middle dose of 1% group,2% group,and the low doses of 1% group,2% group was elevated markedly. Conclusion: Danggui Buxue Decoction serum can promote the proliferation and differentiation of bone marrow hematopoietic progenitor cells,and promote the bone marrow nucleated cells DNA proliferation.
