摘要
目的:研究莪术多糖对猪血清所致肝纤维化大鼠的保护作用。方法:大鼠50只腹腔注射异源猪血清(0.4ml/kg,每周3次,持续8周)复制肝纤维化动物模型。随机分成3组:模型对照组、莪术多糖治疗组(200,400mg/kg),持续灌胃30 d,同时平行设置正常大鼠10只作为正常对照组,对照组均ig给予等体积的生理盐水。末次给药后12h,检测血清丙氨酸转氨酶(ALT),谷草转氨酶(AST)和碱性磷酸酶(ALP)的水平以及测定肝匀浆透明质酸(HA),Ⅲ型前胶原(PCIII),层粘连蛋白(LN)的含量。HE染色观察肝组织组织病理学变化;Western blotting法检测肝组织α-平滑肌动蛋白(α-SMA)、转化生长因子β1(TGFβ1)的表达。结果:与模型对照组比较,莪术多糖(200,400mg/kg)以剂量依赖性地降低猪血清所致大鼠肝纤维化血清肝功能酶水平,同时减少肝组织中胶原蛋白的含量,并显著下调内源性α-SMA和TGFβ1的表达水平,从而缓解肝纤维化病情。结论:莪术多糖具有明显的抗肝纤维化作用,其机制可能与抑制肝纤维化大鼠胶原蛋白过表达以及TGF信号通路有关。
Objective: To study the protective effect of rhizoma zedoariae polysaccharides on porcine serum induced hepatic fibrosis in rats. Methods: Fifty healthy SD rats were intraperitoneally injected heterologous porcine serum (0.4mL/kg, three times per week for 8 weeks) for es- tablishing the animal model of liver fibrosis. The tested rats were randomly divided into three groups : model group, rhizoma zedoariae polysacchafides treatment groups (200,400 mg/kg, ig for 30 days), 10 animals in each group. At the same time, 10 normal rats were parallelly added as control group. All control groups were intrapefitoneally given an equal volume of saline. After the last administration 12h, serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) were measured. Liver homogenates of hyaluronic acid ( HA), Ⅲ procollagen (PCⅢ), laminin (LN) contents were determined. Pathological changes of liver tissue were observed using HE staining. Intrahepatic a-smooth muscle actin (α-SMA), transforming growth factor β1 (TGFβ1) expressions were as- sessed using Western blotting assay. Results: Compared with the model control group, rhizoma zedoariae polysaccharides (200,400 mg/kg) dose-dependently reduced liver function enzyme levels of porcine serum induced hepatofibrosis rats (P 〈 0.01 ) ; while the collagen contents were decreased in liver tissue ( P 〈 0.01 ). Meanwhile, endogenous α-SMA and TGFβ1 expression levels were significantly down-regulated ( P 〈0.01 ), thus alleviating the condition of liver fibrosis. Conclusion: Rhizoma zedoariae polysaccharides have significant role of anti-hepatic fibrosis, which the possible mechanisms are related to inhibition of collagen overexpression in hepatic fibrosis rats and TGF signaling pathway.
