摘要
目的探讨硫化氢(H2S)急性中毒大鼠肺组织核因子κB(NF-κB)含量、分布的动态变化及乌司他丁(UTI)对其的干预作用。方法将SD大鼠88只随机分为对照组、H2S组和H2S+UTI组,用免疫组化法观察肺组织NF-κB含量及分布。结果与对照组比较,H2S组2h,6h,12h,24h,48h时间点大鼠肺组织NF-κB蛋白表达均明显升高(P<0.05);与同时间点H2S组比较,H2S+UTI组2h,6h,12h,24h,48hNF-κB蛋白表达均明显降低(P<0.05)。NF-κB蛋白除可在肺泡上皮细胞表达外,还可分布于气道黏膜上皮细胞、炎性浸润细胞(主要是中性粒细胞和巨噬细胞)中。结论 H2S吸入性中毒急性肺损伤大鼠早期肺组织中NF-κB表达增加,而UTI干预治疗能显著降低NF-κB表达。
Objective To investigate the content and its distribution of dynamic change of nuclear factor κB (NF-κB) of lung tissues from hydrogen sulfide (H2 S) acute poisoned rats and the intervention effects of ulinastatin (UTI) on it. Method 88 SD rats were randomly divided into control group , H2 S group and H2 S+ UTI group and the content and distribution of NF-κB in all groups were observed with the method of immunohistochemistry. Results The protein expressions of NF-κB in the lung tissues of rats in the H2S group increased mor significantly than the control group at the time points of 2, 6, 12, 24 and 48 hours (P〈0.05) ; the protein expressions of NF-κB in the H2 S+UTI group were significantly lower than H2 S group at the same time points of 2, 6, 12, 24 and 48 hours (P〈0.05). NF-κB protein could not only express in alveolar epithelial cells, but also distribute in the tracheal epithelial cells and inflammatory infiltration cells (mainly in neutrophils and macrophages). Conclusion The expression of NF-κB in the lung tissue from rats with H2 S inhalation toxicity and acute lung injury increases in the early stage but the treatment of UTI intervention can significantly reduce its expression.
