摘要
目的研究免疫球蛋白(Ig)M在小鼠心肌组织中的表达及其在正常小鼠和心肌纤维化小鼠心肌组织中的定位。方法将C57BL/10小鼠分成3组:正常组、假手术组和实验组(各4只)。对实验组小鼠进行埋泵并灌注血管紧张素Ⅱ,对假手术组小鼠进行埋泵并灌注生理盐水,正常组不处理。灌注14 d后,处死小鼠,应用Western Blot和免疫组织化学法检测IgM的表达。结果在正常小鼠心肌组织和纤维化心肌组织中均检测到IgM蛋白水平的表达。与正常心肌组织对比,心肌纤维化区域的IgM阳性反应减弱,而周围正常心肌组织中IgM阳性反应则加强。结论心肌细胞内有IgM分子的表达,并且其表达与心肌纤维化相关。
Objective To observe the expression and localization of IgM in mice normal myocardium and fibrotic myocardial tissue. Methods C57BL/10 mice were divided into 3 groups: normal group, sham group and test group (4 mice in each group). The test group was injected with angiotensin II while the sham group was injected with saline for 14 d through a micro pump post in the back hypodermal. Then the mice were killed, Western Blot and immunohistoehemistry staining were taken to detect IgM. Results IgM was detected in normal mice heart tissue and fibrotic heart tissue. Comparing the fibrosis group with the sham group, IgM expression were up-regulated in myocardial cells. In the fibrosis area, the staining seems to be disappear. Conclusions IgM is expressed in mice heart tissue and the expression of IgM may related to myocardial fibrosis.
