摘要
为了构建干旱和盐碱共胁迫的玉米cDNA文库,提取YQ7-96品系的雌雄分化期(12片真叶)的混合组织(叶、茎和花芽)总RNA,运用SMART技术构建pDNR-LIB载体的cDNA文库,随机挑选克隆进行表达序列标签(EST)序列的BlastX比对和MIPS归类分析。结果表明:表达频率10的只有1个独立基因,表达频率4以上的有17个独立基因,单拷贝基因有1 693个。在基因注释中发现了4个编码早期干旱诱导蛋白质的EST,在聚类分析后得到一个片段重叠群,命名为EDIP。重叠群与玉米、水稻的早期干旱诱导蛋白质相关基因有较高的同源性,表明在干旱和盐碱共胁迫的玉米EST数据库中,高频表达的基因较少,大部分为单拷贝基因;另外,已筛选出与干旱和盐碱共胁迫相关的玉米EST。
To make the cDNA library of maize under the co-stress of drought and salt and alkali, RNA was extractedfrom the mixed leaf, stem, and bud tissues of YQ7-96 at 12-leaf stage, and the cDNA library in a pDNR-LIB vector wasconstructed by SMART technique. cDNA clones were randomly picked, and the expression sequence tags(ESTs) were ana-lyzed by BlastX and MIPS. There was one gene with the expression frequency of ten, 17 genes with the expression frequen-cy between four and ten, and 1 693 single copy genes. From the gene annotation, four ESTs encoding an early stagedrought condition-induced protein were found, which formed a contig named EDIP that shared high sequence homologieswith early stage drought condition-induced protein related genes in maize and rice. These findings suggested that in the ESTlibrary of maize under the co-stress of drought and salt and alkali, there were mostly single copy genes with very few highexpression frequency genes. In addition, an EST of maizehad been found correlated with the co-stress of drought andsalt and alkali.
