心宁复方抗心肌缺血再灌注损伤保护作用的实验研究

An experimental study of protective effect of Xinning Fufang against myocardial ischemia- reperfusion injury

摘要 目的:从氧自由基损伤角度探讨心宁复方有效部位群心肌保护作用机制。方法:用原代培养的心肌细胞复制SD大鼠乳鼠缺氧/复氧损伤模型,设立空白对照组,缺氧/复氧组,合贝爽注射液组,心宁复方大、中、小剂量组共6个组。各给药组在缺氧期及复氧期分别给药,测定各组心肌细胞活力、乳酸脱氢酶(LDH)、心肌细胞SOD活性、MDA含量。结果:心宁复方能明显提高受损心肌细胞的活力,降低LDH活性表达,增强SOD活性,减少MDA的含量。结论:抑制心肌细胞氧自由基损伤可能是心宁复方心肌细胞保护作用机制之一。 Objective:To investigate the mechanism of the myocardial protection by effective components of Xinning Fufang from the aspect of oxygen free radical injury. Methods : Primarily cultured cardiomyocytes were treated to establish a neonatal Sprague - Dawley rat model of hypoxia/reoxygenation injury. All subjects were divided into blank control group, hypoxia/reoxygenation group, Herbesser injection group, and high - , middle- , and low- dose Xinning Fufang groups. All treatment groups received medication in the hypoxia and reoxygenation stages. The cardiomyocyte viability, lactate dehydrogenase (LDH)activity, myocardial superoxide dismutase (SOD)activity, and malondialdehyde(MDA) content were determined. Resuhs :Xinning Fufang significantly increased cardiomyocyte viability, reduced LDH activity,increased SOD activity, and reduced MDA content. Conclusion:Inhibiting the oxygen free radical injury of cardiomyocytes may be one of the mechanisms of the myocardial protection by Xinning Fufang.
