摘要
以破壁后过60目筛的雨生红球藻藻粉为原料,采用超临界流体技术提取其中的精油并皂化甲酯化。采用气-质联用仪对其进行定量和定性分析,共鉴定出23种脂肪酸,其中不饱和脂肪酸占总脂肪酸的71.53%,脂肪酸组成主要为棕榈酸、油酸、亚油酸、反亚油酸、α-亚麻酸。37种标准脂肪酸在线性范围内,相关系数均大于0.99。6份平行样品中各种脂肪酸甲酯的RSD为1.79%-9.55%,不同水平下的样品加标回收率范围为73.44%-132.42%,各种脂肪酸甲酯的仪器检测限〈0.65 ng。本文探讨了脂肪酸甲酯在弱极性色谱柱上保留时间的规律。该方法操作简单、迅速、灵敏、准确度高,可满足雨生红球藻及类似性质的藻类脂肪酸的检测需要。
First broken over a 60-mesh sieve Haematococcus pluvialis powder as raw material, The Supercritical fluid extraction CO2 (SFE-CO2) technique extraction was used in the pretreatment of Haematococcus pluvialis, then the fatty acids were converted acid methyl esters, the composition of fatty acid methyl esters was analyzed with gas chromatography and gas chromatography/mass spectrometry. Twenty three fatty acids were found and the unsaturated fatty acidswere 71.53% of total, the main fatty acids were palmitic acid, oleic acid, linoleic acid, trans-linoleic acid, and a- linolenic acid. 37 kinds of standard fatty acids within the linear range, correlation coefficient greater than 0.99. The relative standard deviations of six parallel samples were 1.79%-9.55%. The recoveries for spiked standards in samples were 73.44%-132.42%. The limits of detection for instrument were below 0.65 ng. The rule of retention time for fatty acid methyl ester was investigated. This method is very simple, quick and accurate and could be used in practical application.
