逆转录病毒载体介导的HSVtk基因联合5-FU治疗实验性大肠癌 被引量:10

摘要 目的研究体内外联合应用HSVtk/GCV系统和5-FU治疗大肠癌的疗效。方法用逆转录病毒转染人大肠癌LoVo细胞,MTT法观察GCV联合5-FU对LoVo细胞的杀伤作用。通过裸鼠皮下注射HSVtk+LoVo细胞建立人大肠癌肿瘤模型,应用GCV和(或)5-FU治疗后测量肿瘤体积计算肿瘤生长抑制率,并用HE染色观察有无器官毒副作用。结果体外实验MTT法显示HSVtk/GCV系统联合5-FU对LoVo细胞的杀伤作用较GCV组明显增强(P<0.05),体内实验表明联合治疗组肿瘤体积为(348±68)mm^3大小,明显小于其他各组(P<0.01),HE染色未观察到各器官病理改变。结论联合应用HSVtk/GCV系统和5-FU能提高HSVtk/GCV系统治疗大肠癌的疗效,且没有明显的毒副作用。 AIM To study the effect of synergistic use of herpes simplex virus thymldlne kinase/ganciclovir (HSVtk/GCV) and 5-FU for the treatment of experimental colorectal cancer (CRC). METHODS Recombinant retroviruses containing HSVtk gene were used to infect human colon carcinoma cell line LoVo, HSVtk+/LoVo cells were gelned. The effect of synergistic use of HSVtk/GCV and 5-FU for the treatment of HSVtk+/LoVo cells in vitro was evaluated using MTT assay. We eatabllshed the CRC animal model by subcutaneously injecting HSVtk+/LoVo cells In the nude mlce. After treatment with GCV or (and) 5-FU, the tumor volumes were measured. The side effects were observed with HE staining of organs. RESULTS The effect of synergistic use of HSVtk/GCV and 5.FU for the treatment of HSVtk+/LoVo cells in vitro was better than other groups (P<0.05). In the group treated with HSVtk/GCV and 5-FU, tumor volumes were (348±68mm^3), which was significantly smaller than in other groups. No pethologic changes were observed in organs. CONCLUSION The synerglstlc use of HSVtk/GCV system and 5-FU could Improve the therapeutlc effect for the treatment of CRC.
