摘要
目的:观察痰热清注射液对脓毒症小鼠免疫系统细胞因子的影响,并探讨其作用机制,方法:采用盲肠结扎并穿刺术(CLP)制作小鼠脓毒症模型,将成年雄性BALB/c小鼠随机分为6组:假手术组,CLP手术组、阳性药物组(甲基强的松龙),痰热清大、中、小剂量组。以流式细胞微球阵列法检测外周血细胞因子水平,收集样本后计数胸腺和脾脏淋巴细胞,测定T及B淋巴细胞的增殖功能。结果:CLP导致模型小鼠血清TNF-α、IL-1β、IL-6表达水平明显上升,小鼠胸腺和脾脏萎缩明显,淋巴细胞计数和T及B淋巴细胞的增殖功能均下降,而痰热清和甲强龙处理后能有效减少炎性细胞因子反应;缓解胸腺和脾脏萎缩,3种剂量痰热清均对胸腺及脾脏淋巴细胞的超敏反应有不同程度的阻抑作用,直接刺激T,B淋巴细胞的增殖。结论:痰热清能够有效阻抑脓毒症状态下免疫系统的炎症反应,对脓毒症小鼠有明显的保护作用。
Objective To observe the effect of immune functiom in sepsis mice and to study the mechanism. Methods: Themodels of sepsis was created with Cecalligation and puncture (CLP) and adult male BALB/c mice were divided randomly into 6groups, Sham operation group, CLP group, group with positive drug (Methylprednisolone), Tan Re Qing injection large, mediumand small doses group. The inflammatory cytokines in the serum were detected by the methods of cytometric bead array. Speci-mens were collected to examine the lymphocyte count in the thymus and spleen and test T and B lymphocyte multiplicationfunction,Results: The levels of TNF-ct, IL-6 and IL-10 in the blood were dramatically elevated after CLP. The thymus andspleen of the mice significantly atrophied; both the lymphocyte count and the T and B lymphocyte multiplication function arereduced. After CLP, TRQ and Methylprednisolone had evident effect on inhibiting the increase of inflammatory cytokine, andthose drugs remarkably enhanced the indexes thymus and spleen. Both thymus and spleen lymphocyte count also markedly in-creased in the three groups of TRQ medicine and hymus and spleen lymphocyte multiplication function increased. Conclusion:TRQ could effectively inhibit inflammatory reaction of the immune system caused by sepsis and protect 1 sepsis mice.
