摘要
目的:探讨血必净注射液对脓毒症大鼠肝损伤以及肝组织NF-κB基因表达的影响。方法:40只大鼠随机分成4组:正常对照组(n=10)、假手术组(n=10)、模型组(n=10)、血必净组(n=10)。各组动物分别于术前3d给予血必净注射液5mL/kg或生理盐水5mL/kg尾静脉静推。盲肠结扎穿刺法(CLP)建立脓毒症动物模型。假手术组仅开腹翻动盲肠后关腹。全自动生化分析仪测定血清ALT和AST水平,ELISA检测血清TNF-α水平,Real-time PCR检测肝组织NF-κB基因表达量。结果:与正常对照组相比,模型组大鼠肝功能明显恶化,外周血TNF-α水平明显升高,肝组织NF-κB基因表达量增加;血必净注射液干预后脓毒症大鼠肝功能好转(P<0.05),外周血TNF-α水平降低(P<0.05),肝组织NF-κB基因表达量减少(P<0.05)。结论:血必净注射液可能通过抑制肝组织NF-κB基因的表达而改善脓毒症大鼠的肝功能。
Objective To explore the effect of xuebijing injection on the liver injury and gene expression of nuclear factor kappa B （NF-κB） in septic rats. Methods 40 male rats were randomly divided into control group（ n = 10）,sham operation group（ n = 10）, model group（ n = 10） and xuebijing group（ n = 10）.Before operation,the animals in all the groups were immediately given xuebijing 5mL/kg or physiological saline 5mL/kg intravenous push to caudal vein respectively for 3 days. The sepsis animal models were established with cecum ligation puncture method. The sham operation group was done the same anesthesia and midline laparotomy, the cecum was exteriorized and returned to the abdominal cavity. The levels of ALT and AST in serum were detected with automatic biochemistry analyzer, and TNF-αlevel were detected with enzyme linked immunosorbent assay （EIASA）,NF-κB gene expression were determined by Real-time PCR .Results The the levels of ALT、 AST、TNF-αand the gene expressions of NF-κB significantly increased in model group as compared to control group （P 〈0.05）. Xuebijing injection markedly decreased the levels of ALT , AST、 TNF-αand the gene expressions of NF-κB （P 〈0.05）.Conclusion Xuebijing injection was efficient to ameliorate liver dysfunction in septic rats and effectively lessen liver inflammatory reaction. The mechanism probably by its inhibition of gene expression of NF-κB
