摘要
目的利用siRNA转染干扰沉默ATP结合盒式G型亚家族成员2(ABCG2)基因,并观察沉默前后T24恶性生物学行为的变化。方法 siRNA沉默ABCG2后,观察T24增殖、克隆形成、迁移、侵袭、对放化疗抵抗、成干细胞球能力的影响。结果 siRNA能够使ABCG2 mRNA和蛋白质的表达下降80%左右,ABCG2被下调后,T24的增殖能力、克隆形成能力、迁移和侵袭能力、对放化疗的抵抗能力、成干细胞球的能力均下降(P<0.05)。结论沉默ABCG2能够降低膀胱癌T24的恶性及干性,可以作为一个潜在的治疗靶点。
Objective To investigate the effect on malignant behavior of bladder cancer T24 cells after ABCG2 gene was silenced. Methods After silencing ABCG2 gene by siRNA, we observed the changes of the ability of proliferation, colony formation, mobility and invasion, radiation and chemotherapy resistance, and sphere forming. Results The silent efficience of siR- NA for ABCG2 reached 80%. After downregulating ABCG2, the ability of proliferation, colony formation, mobility and invasion, radiation and chemotherapy resistance, and sphere forming decreased. Conclusion Downregulated ABCG2 can reduce the malignancy and stem cell-like characteristic of T24 bladder cancer cells, and ABCG2 is a potential theraputic target.
