摘要
目的对比不同移植方式移植人脐带间充质干细胞(hUC-MSCs)对成体神经干细胞的原位激活,确定最佳移植途径。方法正常大鼠60只,随机分为对照组、静脉、动脉、蛛网膜下腔移植组。每组15只,各实验组移植数量相同的hUC-MSCs,对照组以假手术组为对照,移植后1、3、7d,利用免疫组织化学方法检测脑组织中BrdU、Nestin的表达。结果对照组BrdU无表达,实验组hUC-MSC在脑内均有BrdU细胞表达,不同途径在1d、3d、7d表达差异有统计学意义(P均<0.001),蛛网膜下腔途径表达最明显;不同途径随时间表达无统计学差异(P>0.05)。Nestin在各组均有细胞表达,分别在1d、3d、7d三个时间点组所测结果与各相应时间点对照组相比(P均<0.001),蛛网膜下腔途径表达最明显;对照组不同途径随时间表达无统计学差异(P均>0.05),各实验组随时间表达差异有统计学意义(P均<0.001)。结论 hUC-MSCs可能对内源性神经干细胞有激活作用,并对神经细胞具营养作用;hUC-MSCs经静脉、动脉、蛛网膜下腔三种途径移植,蛛网膜下腔入路可能是最有效的途径。
Objective To compare the different transplantation,hUC-MSCs the adult NSC situ activation,to determine the best migration path.Methods 60 normal rats were randomly divided into control group,vein,artery,subarachnoid transplantation group (n =15),the hUC-MSCs transplantation the same number of each experimental group,the control group,sham operation group control,day after transplantation,3 d,7 d,text method in which using immunohistochemistry to detect brain tissue in BrdU Nestin expression.Results In the control group,the BrdU expression the experimental group Huc-MSC in the brain had BrdU cells expressing different ways in 1 d,3 d,7 d expression difference was significant(P <0.001),the subarachnoid channels to express the most obvious;Expression had no significant difference(P >0.05) different ways over time.Nestin expression cells in each group,in 1 d,3 d,7 d three time points group measured compared to the results with the corresponding time points in control group(P <0.001),subarachnoid way to express the most obvious; control set of different ways to express no significant difference(P > 0.05) over time,in each experimental group expressed over time the difference was significant (P <0.001).Conclusion hUC-MSCs may be activated endogenous NSC,and nerve cells with nutrition; hUC-MSCs through the veins,arteries,the subarachnoid three ways transplantation,the subarachnoid approach may be the most effective way.
