摘要
为分析生长激素(growth hormone,GH)基因和胰岛素样生长因子1(insulin-like growth factor-1,IGF-1)基因对绵羊生长发育的作用,以6月龄萨福克公羊为试验材料,利用荧光定量PCR SYBR Green I荧光染料法,对绵羊肠系膜淋巴结、心脏、肝脏、垂体、大脑、肾脏、骨骼肌、皮肤、肺脏、睾丸和脾脏11个组织中GH和IGF-1基因的表达水平进行相对定量分析。结果表明,GH基因在睾丸、肠系膜淋巴结和脾脏中的表达量显著高于心脏、肝脏、大脑、肾脏和骨骼肌(P<0.05);IGF-1基因在肝脏和肠系膜淋巴结中的表达量显著高于心脏、垂体、大脑、肾脏、骨骼肌、皮肤、肺脏、睾丸和脾脏(P<0.05)。说明绵羊GH和IGF-1基因有特定的组织表达模式;GH基因与免疫、繁殖及机体的生长发育密切相关;IGF-1基因与生长发育及免疫密切相关,肝脏不仅是IGF-1的主要合成器官,也是该基因特异性表达的主要器官组织之一。GH和IGF-1基因均具有一因多效作用。
To study the role of growth hormone gene(GH)and insulin like growth factor-1 gene (IGF-1)in the growth and development of sheep,the relative expressions of GH and IGF-1 genes were analyzed by real time quantitative PCR with SYBR Green Ⅰ in mesenteric lymph nodes,heart,liver,pituitary,brain,kidney,skeletal muscle,skin,lung,spleen,testes of with 6-month-old male Suffolk sheep.The results showed that IGF-1 gene expression was the highest in the liver,significantly higher than that in the heart,pituitary,brain,kidney,skeletal muscle,skin,lung,testis and spleen(P<0.05); GH gene expression was the highest in the spleen,significantly higher than in that the heart,liver,brain,kidney and skeletal muscle(P<0.05).The results suggested the expression characteristic of GH and IGF-1 gene presented specific developmental patterns,and GH gene was related to the immunity,reproduction,growth and development of sheep body.IGF-1 gene was closely related to the immunity,growth; IGF-1 was mainly secreted in liver,one of the organs for gene specificity expression.GH and IGF-1 gene had a pleiotropic effect.
