摘要
目的 :探讨薏苡仁酯 (Coixenolide,CXL)对平阳霉素 (Pingyangmycin,PYM)抑制人鼻咽癌细胞 CNE- 2 Z增殖的影响。方法 :采用微量细胞克隆形成法检测 CNE- 2 Z对药物的敏感性。结果 :PYM呈剂量依赖性抑制 CNE- 2 Z的增殖 ,ID5 0 =10 - 5 .976 ± 10 - 6 .75 4 mol/L。加入 CXL10 - 7mol/L 和 10 - 6 mol/L均可使 PYM的量效曲线左移 ,ID5 0 降至 10 - 6 .31 5 ±10 - 7.0 5 6 mol/L 和 10 - 6 .71 7± 10 - 7.42 0 mol/L。CXL 还能使 PYM的时效曲线左移 ,缩短该药的作用时间。未发现 CXL 能增强 PYM的急性毒性作用。结论 :CXL选择性地增强 PYM的细胞毒性作用。
Objective:To Examine the feasible influence of coixenolide (CXL) on the antitumour effect of pingyangmycin (PYM,i.e.bleomycin A 5) on human nasopharyngeal carcinoma cell line CNE 2Z. Methods:Micro colony formation assay was used to test the sensitivity of CNE 2Z to PYM.Results:PYM inhibited the proliferation of CNE 2Z in a dose dependance manner,with an ID 50 =10 -5.976 ±10 -6.754 mol/L. Addition of CXL (10 -7 mol/L and 10 -6 mol/L) shifted both the dose effect curve and time response curve of PYM to the left,reducing the ID 50 of PYM to 10 -6.315 ±10 -7.056 mol/L and 10 -6.716 ±10 -7.420 mol/L,respectively,and shortening the duration of PYM's action CXL,however,did not potentiate the acute toxicity of PYM to normal mouse.Conclusion:CXL selectively enhances PYM cytotoxicity to CNE 2Z.
