安胰颗粒对重症急性胰腺炎大鼠NF-κB活化及细胞因子的影响 被引量:4

Effect of Anyi Granules on the Expression of NF-κB Activation and Cytokines in Rats with Severe Acute Pancreatitis

摘要 目的:观察安胰颗粒对重症胰腺炎大鼠NF-κB的影响及其与细胞因子的关系,探讨其对SAP的治疗作用机制。方法:将120只大鼠分为正常组(N组)、模型组(SAP组)、安胰颗粒组(A组)和IL-10组(I组)共4组,各组又分为3小时、6小时、12小时三个亚组。予腹腔内注射6%的左旋精氨酸(L-Arginine)溶液建立SAP大鼠模型。I组予腹腔注射10000U人重组白介素-10(rhIL-10)预处理后诱导SAP。A组连续3天予安胰颗粒4g/kg灌胃后诱导SAP。观察胰腺组织病理学变化,免疫组化检测胰腺组织NF-κBp65的表达,酶联免疫吸附法(ELISA)检测血清TNF-α、IL-1β水平。结果 (1)A组及I组病理学评分较SAP组明显下降(P<0.05)。(2)SAP组胰腺组织NF-κBp65各时点表达显著高于N组(P<0.01);A组NF-κBp65各时点表达较SAP组明显下降(P<0.05),12小时时点表达明显低于I组(P<0.05)。(3)与SAP组相比,A组和I组TNF-α及IL-1β浓度明显下降(P<0.05或P<0.01)。(4)胰腺组织NF-κB p65的表达量与血清TNF-α、IL-1β浓度呈明显正相关关系(P<0.05或P<0.01)。结论:安胰颗粒通过抑制NF-κB活化,减少细胞因子的产生和释放,进而改善胰腺损伤,减轻SAP的炎症反应,有效治疗SAP。 Objective: To observe the effect of AnYi Granules on the expression of NF-κB activation and its relationship with cytokine, which to investigate the mechanism of action for the treatment of SAP. Methods : 120 rats were divided into normal group (N group), model group (SAP group), AnYi pancreas Granules group (A group) and IL-10 group (I group) of four groups, each group was divided into 3h, 6h and 12h three subgroups. The rat model of SAP were established by lumbar injection with L-arginine solution of 6%. I group was used recombinant human interleukin-10 (rhIL-10) of 10000U for pretreatment by intraperitoneal injection and then induced SAP. A group was induced SAP after gavaging AnYi Granules solution of 4g/ kg for three consecutive days. Pathology of pancreatic tissue were observed; the expression of NF-κBp65 in pancreatic tissue were detect by immunohistochemistry; the levels of TNF-α, IL-1β in serum were detect by enzyme-linked immunosorbent assay (ELISA). Results: (1) The pathological score of A group and I group decreased significantly compared with the SAP group (P 〈 0.05). (2) The levels of NF-κBp65 on pancreatic tissue in the SAP group at each time point was significantly higher than the N group (P 〈 0. 01 ) ; the expression of NF-κBp65 of A group at each time point was significantly decreased than the SAP group (P 〈 0. 05), 12h point was significantly lower than the I group (P 〈 0.05). (3) Compared with the SAP group, TNF-α and IL- 1β levels of the A group and the I group were significantly decreased ( P 〈 0. 05 or P 〈 0. 01 ). (4) The expression of NF-κB p65 in Pancreatic tissue and TNF-α, IL-1β concentration in serum was positively correlated (P 〈 0. 05or P 〈 0. 01 ). Conclusion: AnYi Granules can reduce the production and release of cytokines by inhibiting NF-κB activation, thus improve the pancreatic injury, reduce inflammation of SAP which can effectively treat SAP.
